AB  - Prethodne studije su pokazale da klotho poseduje anti-inflamatornu, antifibrotičku i antioksidativnu ulogu, i da postoji direktna pozitivna korelacija izmedu klotho-a i albuminurije kod nekih animalnih modela hronične bubrežne bolesti (CKD). Fokalno segmentna glomeruloskleroza (FSGS) je oblik CKD, koji se odlikuje prisutnom proteinurijom, hipoalbuminemijom i oksidativnim stresom. Ranije smo pokazali da blokada receptora angiotenzina II tipa 1 losartanom (L) smanjuje proteinuriju, glomerulosklerozu, tubulointersticijsku inflamaciju i fibrozu u animalnom modelu FSGS izazvane adriamicinom (ADR, antitumorski lek) kod spontano hipertenzivnih pacova (SHR), ali ove promene u bubrezima su i dalje bile veće od kontrolnih vrednosti. Ekstrakt lista masline (Olea europaea L.) (OLE), dobro poznat kao prirodni antioksidans, bogat je fenolnim jedinjenjima koja se smatraju odgovornim za snažna antioksidativna i anti-inflamatorna dejstva ekstrakta. Stoga smo ispitivali da li tretman losartanom u kombinaciji sa ekstraktom lista masline može biti efikasniji u usporavanju progresije FSGS. Odrasle ženke SHR su podeljene u četiri grupe. Kontrolni pacovi su primili fiziološki rastvor. SHADR, SHADR+L i SHADR+L+OLE grupe su primile ADR (2 mg/kg telesne mase i.v.) dva puta u intervalu od 3 nedelje. Nakon druge injekcije, SHADR+L i SHADR+L+OLE grupe su primale gavažom L (10 mg/kg/dan), odnosno L+OLE (10+80 mg/kg/dan). Na kraju 6-nedeljnog tretmana analizirali smo albuminuriju, nivo azot monoksida (NO,), 6-nitrotriptofan (marker nitracije proteina), klotho i histopatologiju bubrega. U model grupi uočeno je mačajno povećanje albuminurije i indeksa oitećenja bubrega, ito je bilo praćeno značajno smanjenom ekspresijom klotho proteina i 6-nitortriptofana, kao i nivoa NO, u homogenatu bubrega u poredenju sa ovim vrednostima u kontrolnoj grupi. Tretman losartanom je mačajno smanjio albuminuriju, glomerulosklerozu i tubulointersticijska oštećenja u poredenju sa model grupom, ali su oštećenja bubrega i dalje bila mačajno veća nego u kontrolnoj grupi. Takode, vrednost NO, je vraćena na kontrolni nivo, a proteinska ekspresija 6-nitrotriptofana je mačajno povećana, dok je klotho-a ostala nepromenjena, u poredenju sa model grupom. Interesantno, kada je tretmanu losartanom dodat ekstrakt lista masline primećeno je mačajno i još jače smanjenje albuminurije, glomeruloskleroze, tubulointersticijske inflamacije i fibroze u poredenju sa model grupom. Takode, ovi parametri su sniženi do nivoa koji se nisu mačajno razlikovali od onih u kontroli. Pored toga, ekspresija proteina klotho-a u bubrezima je mačajno povećana u poredenju sa grupama SHADR+L i model Naši rezultati su pokazali da je ekstrakt lista masline kombinovan sa losartanom bio efikasniji od samog tretmana losartanom u usporavanju progresije FSGS kod SHR, a ovaj povoljan efekat uključivao je Klotho signalni put.
AB  - Previous studies have shown the anti-inflammatory, antifibrotic and antioxidant properties of klotho, and a direct positive correlation between klotho and albuminuria in some chronic kidney disease (CKD) animal models. Focal segmented glomerulosclerosis (FSGS), a form of CKD, is distinguished by proteinuria, hypoalbuminemia, and oxidative stress. We previously showed that blockade of angiotensin Õ type 1 receptor with losartan (L) decreased proteinuria, glomerulosclerosis, tubulointerstitial inflammation, and fibrosis in animal model of FSGS induced by Adriamycin (ADR, anti-cancer agent) in spontaneously hypertensive rats (SHR), but these renal alterations were still higher than control values. Olive (Olea europaea L.) leaf extract (OLE), well-known as natural antioxidant, is rich in phenolic compounds that are thought to be responsible for its powerful antioxidant and anti-inflammatory properties. Here, we examined if losartan treatment in combination with olive leaf extract consumption could be more effective in slowing down the progression of FSGS. Adult female SHRs were divided in four groups. Control rats received vehicle. SHADR, SHADR+L and SHADR+L+OLE groups received ADR (2 mg/kg body weight i.v.) twice in 3-week-interval. After the second injection, SHADR+L, and SHADR+L+OLE groups received by gavage L (10 mg/kg/day), and L+OLE (10+80 mg/kg/day), respectively. At the end of6-week treatment, we analysed albuminuria, renal nitric oxide level (NO,), 6-nitrotryptophan (marker ofnitration of proteins), klotho, and renal histopathology In model group we found significantly increased albuininuria and renal injury score, followed by significantly decreased klotho protein expressions, NOx level, and 6- nitortryptophan in comparison to these values in control group. Losartan treatment significantly reduced albuminuria, glomerulosclerosis, and tubulointerstitial injuries compared to model group, but renal alterations were still significantly higher than in control group. Furthermore, NO, level reversed to control and 6-nitrotryptophan protein expression significantly increased, but klotho remained unchanged, compared to model group. Interestingly, when olive leaf extract was added to losartan treatment, a significant and even stronger decrease of albuminuria, glomerulosclerosis, tubulointerstitial inflammation and fibrosis were observed in comparison to model group. Also, these parameters were lowered to the levels not significantly different than in control. Furthermore, renal protein expression of klotho was significantly increased compared to SHADR+L and model groups. Our results showed that olive leaf extract in addition to losartan treatment was more effective than losartan alone in slowing down the progression of FSGS in SHR, and that beneficial effect involved the klotho signalling pathway.

AB  - In order to exploit the advantage of drug delivery through the buccal mucosa, mucoadhesive buccal films with propranolol hydrochloride based on polyethylene oxide, hydroxypropyl methylcellulose, and polyvinyl alcohol have been developed. The aim of this study was the development, pharmaceutical-technological (uniformity of mass and drug content, film thickness, potential interactions between polymers and propranolol hydrochloride, in vitro drug permeation and drug release, mechanical and mucoadhesive properties), and biopharmaceutical characterization of prepared mucoadhesive buccal films through conducting in vivo study in spontaneously hypertensive rats and in silico modeling of intraoral and gastrointestinal drug absorption. For in vivo study formulation F2 (hydroxypropylmethyl cellulose 0.5%, polyethylene oxide 3.5%, polyvinyl alcohol 1.5%, propylene glycol 3%, and propranolol hydrochloride 2%) was selected, which showed the highest values of tensile strength and percentage of elongation, as well as the highest value of the force of adhesion. Results of pharmacokinetic in rats and in silico modeling confirmed the superiority of the mucoadhesive buccal films over immediate-release tablets (in rats: AUC0→24h 66.13 ± 18.03 vs. 24.61 ± 5.52 μg⋅h/mL, AUC0→∞ 111.82 ± 39.04 vs. 47.85 ± 11.67 μg⋅h/mL; in silico: AUC0→24h 200.17 vs.74.58 ng⋅h/mL, AUC0→∞ 204.04 vs. 75.64 ng⋅h/mL). Hemodynamic measurements have shown that mucoadhesive buccal films, compared to immediate-release tablets, provide a more pronounced decrease primarily in heart rate (28-51%), but also in diastolic pressure (up to 33%), as well as a longer heart rate reduction that was maintained for up to 12th h. Therefore, mucoadhesive buccal films increased the degree of absorbed drug and thus contributed to better therapeutic outcomes compared to immediate-release tablets for peroral administration.

AB  - Hipertenzija, koja se izmedu ostalog karakteriše i deficitom vasodilatatornog azotnog monoksida
(AM), mačajno pogoršava kliničku sliku postishernične akutne bubrežne slabosti (ABS) kod ljudi. Ponovno vraćanje krvi u bubrežno tkivo, samo po sebi dovodi do masovne produkcije slobodnih vrsta kiseonika (SVK) i pogoršanja ishemijske povrede. Ispitivali smo potencijalni zaštitni efekat primene donora AM-a, kao i uklanjanja SVK na sistemske i bubrežne hemodinamske parametre, kao i na strukturu i funkciju bubrega pacova sa urodenom hipertenzijom (PUH), kojima je izazivana postishemična ABS. Za navedene potrebe, PUH su dobijali donor AM, L-arginin, prirodni sakupljač SRK vitamin C (Vit C) ili sintetski mimetik enzima superoksid dismutase TEMPOL, tokom ishemije-reperfuzije bubrega u trajanju od 40 minuta. Tretman L-argininom je poboljšao sistemsku i bubreżnu hemodinarniku, ali nije uspeo da smanji ośtećenje bubrega. Nasuprot tome, intenzitet nekroze i dilatacije tubula su bili mačajno smanjeni kod pacova u ABS+Vit C grupi u poredenju sa kontrolnom ABS grupom. Uprkos smanjenju lipidne peroksidacije u plazmi, kao i renalnog vaskularnog otpora, tretman TEMPOL-om nije uspeo da ublaži reperfuzijom nastala oštećenja bubreżnog tkiva kod PUH. Naši rezultati ukazuju na to da SVK učestvuju u reperfuzionim oštećenjima bubrega, a da su sintetski mimetik superoksid dismutaze i donor AM neefikasni u zaštiti glomerula i tubula nakon nastanka ABS kod PUH. Sa druge strane, naši rezultati sugerišu daje Vit C, prirodni antioksidant, hemoprotektant umerenog zaśtitnog dejstva protiv ishemijsko-reperfuzionih povreda bubrega u hipertenzivnim okolnostima. Ova zaštita je većinski povezana sa direktnim uklanjanjem SVK.
AB  - Hypertension, which is among others characterized by reducing of nitric oxide (NO) mediated vasodilatation, strongly aggravates postischemic acute kidney injury (AKI) in humans. Kidney reperfusion itself increases ischemic injury through the massive production of oxygen-free radicals. We investigated protective potential of NO supplementation and superoxide radical scavenging on systemic and renal haemodynamic parameters as well as kidney function and structure in spontaneously hypertensive rats (SHR) with induced postischemic AKI. For these purposes, SHR were subjected to NO donor L-arginine, natural oxidant scavenger vitamin C (Vit C) or synthetic superoxide dismutase mimetic TEMPOL during 40 minutes kidney ischaemia-reperfusion period. L-arginine supplementation improves systemic and renal hemodynamic but failed to reduce kidney tubular injuries. In opposite, intensity of tubular necrosis and dilatation was markedly reduced in AKI+Vit C group in comparison to AKI control. Despite blunting of lipid peroxidation in plasma and diminishing of renal vascular resistance, TEMPOL treatment also failed to reduce kidney tissue injuries induced by reperfusion in SHR. Our results suggest that superoxide radicals participate in kidney reperfusion injuries while synthetic superoxide dismutase and NO supplementation are ineffective in glomerular and tubular protection after AKI episode in hypertensive rats. On the other hand, our results promote natural antioxidant, vitamin C, as a moderately effective chemoprotectant against kidney ischemia-reperfusion injuries in hypertensive surrounding. Protection is mainly related to the direct superoxide radical scavenging.

Ponovno vraćanje krvi u bubrežno tkivo, samo po sebi dovodi do masovne produkcije slobodnih vrsta kiseonika (SVK) i pogoršanja ishemijske povrede. Ispitivali smo potencijalni zaštitni efekat primene donora AM-a, kao i uklanjanja SVK na sistemske i bubrežne hemodinamske parametre, kao i na strukturu i funkciju bubrega pacova sa urodenom hipertenzijom (PUH), kojima je izazivana postishemična ABS. Za navedene potrebe, PUH su dobijali donor AM, L-arginin, prirodni sakupljač SRK vitamin C (Vit C) ili sintetski mimetik enzima superoksid dismutase TEMPOL, tokom ishemije-reperfuzije bubrega u trajanju od 40 minuta. Tretman L-argininom je poboljšao sistemsku i bubreżnu hemodinarniku, ali nije uspeo da smanji ośtećenje bubrega. Nasuprot tome, intenzitet nekroze i dilatacije tubula su bili mačajno smanjeni kod pacova u ABS+Vit C grupi u poredenju sa kontrolnom ABS grupom. Uprkos smanjenju lipidne peroksidacije u plazmi, kao i renalnog vaskularnog otpora, tretman TEMPOL-om nije uspeo da ublaži reperfuzijom nastala oštećenja bubreżnog tkiva kod PUH. Naši rezultati ukazuju na to da SVK učestvuju u reperfuzionim oštećenjima bubrega, a da su sintetski mimetik superoksid dismutaze i donor AM neefikasni u zaštiti glomerula i tubula nakon nastanka ABS kod PUH. Sa druge strane, naši rezultati sugerišu daje Vit C, prirodni antioksidant, hemoprotektant umerenog zaśtitnog dejstva protiv ishemijsko-reperfuzionih povreda bubrega u hipertenzivnim okolnostima. Ova zaštita je većinski povezana sa direktnim uklanjanjem SVK., Hypertension, which is among others characterized by reducing of nitric oxide (NO) mediated vasodilatation, strongly aggravates postischemic acute kidney injury (AKI) in humans. Kidney reperfusion itself increases ischemic injury through the massive production of oxygen-free radicals. We investigated protective potential of NO supplementation and superoxide radical scavenging on systemic and renal haemodynamic parameters as well as kidney function and structure in spontaneously hypertensive rats (SHR) with induced postischemic AKI. For these purposes, SHR were subjected to NO donor L-arginine, natural oxidant scavenger vitamin C (Vit C) or synthetic superoxide dismutase mimetic TEMPOL during 40 minutes kidney ischaemia-reperfusion period. L-arginine supplementation improves systemic and renal hemodynamic but failed to reduce kidney tubular injuries. In opposite, intensity of tubular necrosis and dilatation was markedly reduced in AKI+Vit C group in comparison to AKI control. Despite blunting of lipid peroxidation in plasma and diminishing of renal vascular resistance, TEMPOL treatment also failed to reduce kidney tissue injuries induced by reperfusion in SHR. Our results suggest that superoxide radicals participate in kidney reperfusion injuries while synthetic superoxide dismutase and NO supplementation are ineffective in glomerular and tubular protection after AKI episode in hypertensive rats. On the other hand, our results promote natural antioxidant, vitamin C, as a moderately effective chemoprotectant against kidney ischemia-reperfusion injuries in hypertensive surrounding. Protection is mainly related to the direct superoxide radical scavenging.",

AB  - Ischemia–reperfusion injury (IRI) is a frequent cause of AKI, resulting in vasoconstriction, cellular dysfunction, inflammation and the induction of oxidative stress. DNA damage, including physical DNA strand breaks, is also a potential consequence of renal IRI. The histone H2A variants, primary H2AX and H2AZ participate in DNA damage response pathways to promote genome stability. The aim of this study was to evaluate the immunohistochemical pattern of histone H2A variants’ (H2AX, γH2AX(S139), H2AXY142ph and H2AZ) expression in an experimental model of ischemia–reperfusion-induced acute kidney injury in spontaneously hypertensive rats. Comparing the immunohistochemical nuclear expression of γH2AX(S139) and H2AXY142ph in AKI, we observed that there is an inverse ratio of these two histone H2AX variants. If we follow different regions from the subcapsular structures to the medulla, there is an increasing extent gradient in the nuclear expression of H2AXY142ph, accompanied by a decreasing nuclear expression of γH2AX. In addition, we observed that different structures dominated when γH2AX and H2AXY142ph expression levels were compared. γH2AX was expressed only in the proximal tubule, with the exception of when they were dilated. In the medulla, H2AXY142ph is predominantly expressed in the loop of Henle and the collecting ducts. Our results show moderate sporadic nuclear H2AZ expression mainly in the cells of the distal tubules and the collecting ducts that were surrounded by dilated tubules with PAS (periodic acid–Schiff stain)-positive casts. These findings may indicate the degree of DNA damage, followed by postischemic AKI, with potential clinical and prognostic implications regarding this condition.

AB  - Objective Background: The main cause of death among patients with malignant hypertension is a kidney failure. The promising field in essential and malignant hypertension therapy could be centered on the amelioration of oxidative stress using antioxidant molecules like resveratrol. Resveratrol is a potent antioxidative agent naturally occurred in many plants that possess health-promoting properties. Methods: In the present study, we investigated the therapeutic potential of resveratrol, a polyphenol with anti-oxidative activity, in NG-L-Arginine Methyl Ester (L-NAME) treated spontaneously hypertensive rats (SHR) - malignantly hypertensive rats (MHR). Results: Resveratrol significantly improves oxidative damages by modulation of antioxidant enzymes and suppression of prooxidant factors in the kidney tissue of MHR. Enhanced antioxidant defense in the kidney improves renal function and ameliorates the morphological changes in this target organ. Besides, protective properties of resveratrol are followed by the restoration of the nitrogen oxide (NO) pathway. 4) Conclusion: Antioxidant therapy with resveratrol could represent promising therapeutical approach in hypertension, especially malignant, against kidney damage.

Oxidative stress has been considered as a central aggravating factor in the development of postischemic acute kidney injury (AKI). 4-hydroxynonenal (4-HNE) is a product of lipid peroxidation and may serve as non-invasive biomarker of oxidative stress while neutrophil gelatinase-associated lipocalin (NGAL) is a biomarker of AKI. Hypoxia-inducible factors (HIFs) are the “master’’ transcription factors responsible for gene expression in hypoxia, which play an important role in kidney injury and repair by regulating HIF target genes, including microRNAs.
Design and method: 
The study was conducted on male SHR rats, randomly divided into following groups: sham-operated group (SHAM) AKI control group, AKI group with apocynin treatment (AKI+APO), group with HBO preconditioning before AKI induction (AKI+HBO) and group with HBO preconditioning before and apocynin treatment after AKI induction (AKI+APO+HBO). HBO preconditioning was performed by exposing to pure oxygen (2.026 bar) twice a day for two consecutive days for 60 minutes, and 24 h before AKI induction. Apocynin, NADPH oxidase inhibitor was injected 5 minutes before reperfusion.
Results: 
AKI induction significantly decreased creatinine (CCr), urea (CU) and phosphate (CPhos) clearances when compared to SHAM group. Remarkable increase in CCr, CU, CPhos levels were observed in all treated groups (AKI+APO, AKI+HBO, AKI+APO+HBO) in comparison to AKI group. Sham – operated rats did not express 4-HNE and NGAL in any parenchymal structure. AKI induced abundant and strong glomerular expression of 4-HNE along with expression in interstitial compartment. All treatments (AKI+APO, AKI+HBO, AKI+APO+HBO) significantly decreased 4-HNE expression both in glomeruli and interstitium. Also, AKI stimulated widespread NGAL expression in renal epithelial tubular cells with fine granular appearance on the apical surface of the cells, affecting entire circumference of tubular cross-sectioning, which was decreased in all treated groups. In SHAM group, the expression of HIF1 alpha and HIF2 alpha was focal with weak intensity in glomeruli and interstitium. In AKI group, the expression was diffuse and strong, with remarkable improvement in all treated groups.
Conclusions: 
Considering our results, we can conclude that apocynin treatment, as well as HBO preconditioning may have protective effects in postischemic acute kidney injury induced in spontaneously hypertensive rats.

AB  - Uvod. Reaktivne vrste kiseonika (RVK), važni signalni molekuli, u uslovima prekomerne produkcije dovode do poremećaja funkcije i strukture organa i tkiva, uključujući bubreg. RVK doprinose dijabetičnim nefropatijama, hipertenzijom i ishemijom izazvanim bolestima bubrega, kao i toksičnim nefropatijama. Cilj i metode. Proučavali smo, na eksperimentalnim modelima, uticaj RVK na nastanak i razvoj fokalno-segmentne glomerulo-skleroze i postishemične akutne bubrežne slabosti. Rezultati. Naši rezultati su pokazali štetno delovanje reperfuzijom indukovane produkcije RVK na protok krvi kroz bubrežnu arteriju, glomerusku i tubulsku funkciju bubrega, kao i strukturu bubrežnog tkiva. Sa druge strane, uočili smo brojne interakcije ovih molekula sa azotnim monoksidom, sa kojim formiraju visoko reaktivne peroksinitrite i oštećuju tkivo bubrega. Evidentna je i povezanost RVK sa renin angiotenzin sistemom, jer je blokada receptora za angiotenzin 2 dovela do poboljšanja strukture i funcije bubrega u oba modela nefropatija. Akutno uklanjanje RVK mimeticima superoksid dismutaze, kao i pretretman hiperbaričnim kiseonikom, su takođe dovodili do usporavanja toka i razvoja nefropatija, sa određenim, neočekivanim, odstupanjima u ishodima kombinovane terapije. Zaključak. RVK igraju značajnu ulogu u patofiziološkim mehanizmima nefropatija, a strategije njihovog neutralisanja predstavljaju obećavajući pristup terapiji ovih poremećaja.

AB  - Kidney disease represents a serious global health problem. Free radicals and prooxidants produced during ischemic acute kidney injury (AKI) may further aggravate the course of the disease and play a role in the pathogenesis of subsequent complications. The aims of our study were to examine the importance of NADPH oxidase blockade and to determine the effect of hyperbaric oxygen preconditioning on the immunohistochemical analysis of 4-hydroxynonenal (4-HNE), neutrophil gelatinase-associated lipocalin (NGAL), and heme oxygenase-1 (HO-1) tissue expression in postischemic acute kidney injury induced in spontaneously hypertensive rats (SHR). Twenty-four hours before AKI induction, HBO preconditioning was carried out by exposing to pure oxygen (2.026bar) twice a day, for 60 min in two consecutive days. Acute kidney injury was induced by removal of the right kidney while the left renal artery was occluded for 45 min by atraumatic clamp. NADPH oxidase blockade was performed by Apocynin (40 mg/kg body weigh)t, intravenously, 5 min before reperfusion. We showed increased 4-HNE renal expression in postischemic AKI compared to Sham operated (SHAM) group. Apocynin treatment, with or without HBO preconditioning, improved creatinine and phosphate clearances, in postischemic AKI. This improvement in renal function was accompanied with decreased 4-HNE, while HO-1 kidney expression restored close to the control group level. NGAL renal expression was also decreased after apocynin treatment, and HBO preconditioning,with or without APO treatment. Considering our results, we can say that 4-HNE tissue expression can be used as a marker of oxidative stress in postischemic AKI. On the other hand, NADPH oxidase blockade and HBO preconditioning reduced oxidative damage, and this protective effect might be expected even in experimental hypertensive condition.

AB  - The  aim  of  this  study  was  to  optimize  the  parameters  for  the  extraction  of  total  flavonoids  from stinging nettle leaf. Comparison of the effects of different solvents on total flavonoid content showed that, regardless  of  extraction  time,  aqueous  methanolicextracts  had  higher  total  flavonoid  content  than  did aqueous ethanolic extracts. So, full factorial design and response  surface  methodology (RSM)  were  em-ployed to estimate the effects of methanol content (50, 75and 100%) and extraction time (30, 60and 90 min) on the total flavonoid content and antioxidant capacities of the extracts. RSM analysis showed that methanol  content  in  the  solvent  influenced  significantly  total  flavonoid  content  and FRAP  (ferric-reducing antioxidant power) antioxidant capacity, while extraction time had no significant effect on either of these responses. Extraction parameters for maximal total flavonoid content were estimated to be 69% aqueous  methanol  and  67  min,  and  65%  aqueous  methanol  and  83  min  for  maximal FRAPantioxidantcapacity.  DPPH  (2,2-diphenyl-1-picrylhydrazyl)  antioxidant  capacity  was  not  significantly  affected  by extraction time or methanol percentage in the solvent.

AB  - Hypertension is one of the most prevalent and powerful contributors of cardiovascular diseases. Malignant hypertension is a relatively rare but extremely severe form of hypertension accompanied with heart, brain, and renal impairment. Resveratrol, a recently described grape-derived, polyphenolic antioxidant molecule, has been proposed as an effective agent in the prevention of cardiovascular diseases. This study was designed to examine chronic resveratrol administration on blood pressure, oxidative stress, and inflammation, with special emphasis on cardiac structure and function in two models of experimental hypertension. The experiments were performed in spontaneously (SHRs) and malignantly hypertensive rats (MHRs). The chronic administration of resveratrol significantly decreased blood pressure in both spontaneously and malignant hypertensive animals. The resveratrol treatment ameliorated morphological changes in the heart tissue. The immunohistochemistry of the heart tissue after resveratrol treatment showed that both TGF-β and Bax were not present in the myocytes of SHRs and were present mainly in the myocytes of MHRs. Resveratrol suppressed lipid peroxidation and significantly improved oxidative status and release of NO. These results suggest that resveratrol prevents hypertrophic and apoptotic consequences induced by high blood pressure with more pronounced effects in malignant hypertension.

AB  - Olive (Olea europaea L.) leaf extract (OLE) possesses powerful antioxidant, antihyperlipidemic, and anti-inflammatory properties. The aim was to investigated the effects of OLE on the hyperlipidemia, antioxidant defense, heme oxygenase/biliverdin reductase (HO/BVR) pathway, inflammation, and fibrosis in spontaneously hypertensive rats with focal segmental glomerulosclerosis (FSGS, a progressive form of chronic kidney disease) induced by adriamycin (2 mg/kg, i.v., twice in a 21-day period). Daily treatment of OLE (80 mg/kg, p.o.) for 6 weeks suppressed protein oxidation and lipid peroxidation (p  lt  .01 and p  lt  .001, respectively), significantly increased antioxidant enzymes activities and normalized antioxidant capacity, leading to the improvement of antioxidant defense independently of the HO/BVR pathway. Furthermore, the values of triglycerides (p  lt  .01), total, and low-density lipoprotein cholesterol (p  lt  .05, both) were improved by OLE. OLE strongly prevented glomerulosclerosis, interstitial inflammation, and fibrosis (renal injury score, FSGS: 8 +/- 0.45 vs. FSGS+OLE: 4.20 +/- 1.07; p  lt  .01), as evidenced by normalized fibronectin content (p  lt  .001), suppressed interstitial inflammatory cells infiltration and collagen deposition, without changing cytokines expressions. OLE decreased blood pressure with a tendency to reduce urine albumin loss. These data suggest that OLE may be effective in slowing down the progression of FSGS.

AB  - Renal ischemia and reperfusion (I/R) injury is the most common cause of acute kidney injury (AKI). Pathogenesis of postischemic AKI involves hemodynamic changes, oxidative stress, inflammation process, calcium ion overloading, apoptosis and necrosis. Up to date, therapeutic approaches to treat AKI are extremely limited. Thus, the aim of this study was to evaluate the effects of hyperbaric oxygen (HBO) preconditioning on citoprotective enzyme, heme oxygenase-1 (HO-1), pro-apoptotic Bax and anti-apoptotic Bcl-2 proteins expression, in postischemic AKI induced in normotensive Wistar and spontaneously hypertensive rats (SHR). The animals were randomly divided into six experimental groups: SHAM-operated Wistar rats (W-SHAM), Wistar rats with induced postischemic AKI (W-AKI) and Wistar group with HBO preconditioning before AKI induction (W-AKI + HBO). On the other hand, SHR rats were also divided into same three groups: SHR-SHAM, SHR-AKI and SHR-AKI + HBO. We demonstrated that HBO preconditioning upregulated HO-1 and anti-apoptotic Bcl-2 protein expression, in both Wistar and SH rats. In addition, HBO preconditioning improved glomerular filtration rate, supporting by significant increase in creatinine, urea and phosphate clearances in both rat strains. Considering our results, we can also say that even in hypertensive conditions, we can expect protective effects of HBO preconditioning in experimental model of AKI.

AB  - Lifestyle modifications such as increase in high-fat food consumption importantly increases the risks for cardiovascular disease. The principal objective of this study is to analyze effects of different high fat diet (HFD) sources on haemodynamic parameters, lipid and oxidative profile, myeloperoxidase activity, and markers of inflammation (IL-6/pentraxin-3). HFD containing 20% of fat, provided by lard (saturated) or soybean oil (unsaturated), as well as control diet were administering to three groups (L, SO and C). Food efficiency ratio and plasma lipids were significantly elevated in both HFD groups. However, only SO group showed an increase in systolic arterial pressure, oxidative stress index, myeloperoxidase activity, liver lipids as well as markers of inflammation: IL-6 and pentraxin-3 (PTX3). In summary, these results indicate inflammogenic potential of excessive soybean oil consumption in triggering liver damage.

AB  - Oxidative stress has been considered as a central aggravating factor in the development of postischemic acute kidney injury (AKI). The aim of this study was to perform the immunohistochemical analysis of 4-hydroxynonenal (4-HNE), neutrophil gelatinase-associated lipocalin (NGAL), and heme oxygenase-1 (HO-1) tissue expression after apocynin (APO) treatment and hyperbaric oxygenation (HBO) preconditioning, applied as single or combined protocol, in postischemic acute kidney injury induced in spontaneously hypertensive rats (SHR). Twenty-four hours before AKI induction, HBO preconditioning was carried out by exposing to pure oxygen (2.026 bar) twice a day, for 60 min in two consecutive days. Acute kidney injury was induced by removal of the right kidney while the left renal artery was occluded for 45 min by atraumatic clamp. Apocynin was applied in a dose of 40 mg/kg body weight, intravenously, 5 min before reperfusion. We showed increased 4-HNE renal expression in postischemic AKI compared to Sham-operated (SHAM) group. Apocynin treatment, with or without HBO preconditioning, improved creatinine and phosphate clearances, in postischemic AKI. This improvement in renal function was accompanied with decreased 4-HNE, while HO-1 kidney expression restored close to the control group level. NGAL renal expression was also decreased after apocynin treatment, and HBO preconditioning, with or without APO treatment. Considering our results, we can say that 4-HNE tissue expression can be used as a marker of oxidative stress in postischemic AKI. On the other hand, apocynin treatment and HBO preconditioning reduced oxidative damage, and this protective effect might be expected even in experimental hypertensive condition.

AB  - The objective of this study was to formulate extended-release mucoadhesive buccal tablets of propranolol hydrochloride in order to provide a prolonged absorption of propranolol hydrochloride from the buccal mucosa and to reduce presystemic metabolism and thus provide a better therapeutic effect. Besides, the aim was to perform comparative in vivo pharmacokinetic and hemodynamic studies of the developed extended-release (ER) propranolol hydrochloride 10 mg mucoadhesive buccal tablets and commercial immediate-release (IR) propranolol hydrochloride 10 mg tablets in spontaneously hypertensive rats. Formulation with 15% polyethylene oxide showed the highest degree of propranolol hydrochloride permeation, satisfactory mucoadhesiveness, and extended-release of propranolol hydrochloride, thus it was selected for further in vivo study. The pharmacokinetic study in rats showed the superiority of ER mucoadhesive buccal tablets over IR tablets in terms of propranolol hydrochloride absorption extent (AUC values: 70.32 ± 19.56 versus 31.69 ± 6.97 µg·h/mL), although lower maximum plasma propranolol hydrochloride concentration (Cmax) was achieved. However, no statistically significant difference was observed in Cmax between these treatments. The hemodynamic study showed that ER mucoadhesive buccal tablets provide a more pronounced decrease primarily in heart rate, but also in systolic and diastolic arterial pressure, as well as a longer heart rate reduction compared to IR tablets.

The aim of this study was to determine the baseline damage to DNA, protein, and lipid macromolecules in different organs of spontaneously hypertensive rats. Markers of lipid peroxidation, protein oxidation, and DNA damage were measured in blood, heart, kidney, and liver of 24-week-old spontaneously hypertensive rats. Plasma prooxidant and antioxidant status were determined as well. Age-matched normotensive Wistar rats were used as control. A rise in markers of lipid peroxidation and protein oxidation, malondialdehyde, and advanced oxidation protein products, was detected in all tissues of spontaneously hypertensive rats, with particularly high values in the liver. DNA damage, measured by the comet assay, was significantly higher in all the studied tissues of spontaneously hypertensive rats compared to normotensive control, with more severe damage in the cardiac and renal cells. Significant depletion of the plasma antioxidant barrier in spontaneously hypertensive rats was also observed. This study showed increased damage to all macromolecules in all studied samples of spontaneously hypertensive rats in comparison with control Wistar rats.

Olive leaf extract improves renal antioxidant defense without altering the heme oxygenase-1/biliverdin reductase pathway in hypertensive rats with focal segmental glomerulosclerosis
